This study was conducted to characterize the Bacillus populations associated with dead Rhynchophorus ferrugineus, to develop a biological control for the red palm weevil. Dead adult beetles, collected throughout Sicily, were used for isolating internal and external spore forming bacteria (SFB) microbiota. The isolates, preliminarily allotted to the Bacillaceae family, were tested at 4 concentrations (103 to 106 CFU/mL) for their ability to inhibit hatching of eggs of R. ferrugineus and were used at 106 CFU/mL to monitor their insecticidal activity against 10 day-old larvae. In this study, the composition of the cultivable microbial populations of 38 nectar honey and honeydew honey samples of different botanical and geographical origin were assessed. After growth in specific media, various colonies with different appearance were isolated and purified before phenotypic (morphological, physiological and biochemical traits) and genotypic [randomly amplified polymorphic DNA (RAPD), repetitive DNA elements-PCR (rep-PCR) and restriction fragment length polymorphism (RFLP)] differentiation. Antifungal peptides produced by Bacillus amyloliquefaciens AG1 active against grapevine fungal pathogens

Abstract Antifungal metabolites produced by Bacillus amyloliquefaciens AG1, previously isolated from wood of grapevine with “esca syndrome”, were studied. The crude protein extract (CPE) obtained from culture supernatant fluid by precipitation with ammonium sulfate was assayed against many grapevine fungal pathogens. B. amyloliquefaciens strain AG1 showed a broad spectrum of antifungal activity, inhibiting mycelial growth in vitro of all tested fungal microorganisms. Food bio-preservatives are requested as substituents of chemical pesticides in food. The aim of this study was to carry out a screening of twenty biocontrol agents (BCAs) for their potential fungicidal activity in vitro. Twenty BCAs were tested against ten pathogenic fungi. Some of the cell-free supernatants (CFS) tested showed in vitro antifungal activity versus pathogenic fungi. Preliminary investigations on grapevines (cv Catarratto) with symptoms of “esca” allowed the isolation of different bacterial colonies (Alfonzo et al, 2009). A Gram-positive, spore forming isolate, able to inhibit fungal growth, was subjected to identification. Aspergillus is a widespread fungal genus contaminating and degrading food and feeds mostly during storage. Some species can also produce carcinogenic mycotoxins harmful for human and animal health. In this study, Bacillus amyloliquefaciens AG1 (BaAG1), active against fungal pathogens was evaluated by in vitro tests against seven Aspergillus species belonging to six Aspergillus sections. After 7 days of incubation at 25°C, mycelial growth of all fungal species was reduced from 46.53% to 60.92%, compared to the controls. Pathogenicity and possible biological control of fungi associated to Botryosphaeria dieback of grapevine in Western Sicily

Botryosphaeria dieback is a widespread grapevine trunk disease caused by species of fungi within the family Botryosphaeriaceae. Symptoms of the disease include bud necrosis, leaf chlorosis, vascular discoloration of the wood, cankers and dieback. In 2013 typical symptoms of Botryosphaeria dieback was observed in Western Sicily and four species associated with the syndrome were isolated and identified as Diplodia seriata, Lasiodiplodia mediterranea, Neofusicoccum parvum and Neofusicoccum vitifusiforme.
